※ SCY family introduction

SCY1 family is composed of three members in human genome (1). SCYL1 is involved in regulating COPI-mediated retrograde traffic. Isoform 6 acts as transcriptional activator. It binds to three different types of GC-rich DNA binding sites (box-A, -B and -C) in the beta-polymerase promoter region. It also binds to the TERT promoter region (2). SCYL2 is component of AP2-containing clathrin coated structures at the plasma membrane or of endocytic coated vesicles and may regulate clathrin-dependent trafficking between the TGN and/or the endosomal system (3).
